Doom was considered too dangerous to remain at the university, and left to pursue his destiny, journeying to the Himalayas, where he developed his skills among Tibetan monks, who assisted him in making his metal mask and armor.
Back in Latveria, Doom led a revolution and took control of the country, making himself its leader. From their, his story became a key part of Marvel’s Universe.

In a direct fight Thanos would easily overpower Doctor Doom. Doom might have a shot if he uses his magic, like Doctor Strange did in Avenger: Infinity War on Titan.
